Ciprick says the Notice published by Health Canada’s Office of Legislative and Regulatory Affairs Controlled Substances Directorate was welcomed by Optimi, specifically around the “importance of drug quality and Good Manufacturing Practices (GMP).”

As a tier one ingredient provider and product leader, Optimi’s operational footprint includes 20,000 square feet of state-of-the-art, technologically advanced aeroponics facilities dedicated to the cultivation of EU-GMP grade psilocybin and functional mushrooms.

Optimi Labs Inc., a wholly owned subsidiary of Optimi Health, filed the amendment in March 2022 in order to ensure adequate supply for licensed companies undertaking molecular development research and Phase 2a and 2b clinical trials investigating the treatment of addictions and mental health conditions. The strategy is part of the Company’s goal to commercialize operations in 2022.

The initial batch of Optimi’s natural psilocybin mushrooms is intended specifically for analysis and use in human clinical trials where market demand for natural psilocybin is increasing.

Optimi is involved in a partnership with the IMPACT Clinical Trial Accelerator Program housed within Alberta’s Life Sciences Innovation Hub at the University of Calgary and has opted to assess its product prior to completion of the Company’s clinical trial application.

Optimi has received a Section 56 Exemption to the Controlled Drugs and Substances Act from Health Canada in order to carry out its own research at the premises of Calgary-based Bloom Psychedelic Therapy & Research Centre (“Bloom”), with a focus on determining a safe and effective standardized microdose of psilocybin for mental health conditions such as anxiety and depression.

With a mission to provide compassionate, frontline solutions to Canada’s growing mental health crisis, the Company plans to supply psychedelics like psilocybin to those truly in need.

Announced on January 5, 2022, the SAP allows qualified medical practitioners to request access to Schedule 3 controlled substances such as psilocybin on a case-by-case emergency basis. Prior to the new regulatory amendments, medical practitioners were prohibited from requesting access to psilocybin and MDMA for individual psychedelic-assisted psychotherapy. The amendments have the potential to positively affect the lives of people experiencing serious mental health conditions and end-of-life distress when other therapies have failed, are unsuitable or are unavailable in Canada.

Optimi’s CEO, Bill Ciprick, says the SAP is already having a positive impact on patients in need and will provide the Company with the right avenue to develop and supply the demand for natural psilocybin.

The extraction process uses a highly viscous material to isolate key components of psychedelic mushrooms and dissolves the psilocybin into a molecular solution. Using the same energy, the active ingredients are then repelled into the skin when applied topically. Due to the skin’s highly permeable membrane, Optimi’s solution has a natural time-release effect, which allows for a more sustained release into the body and provides a patient with the ability to experience the desired outcome for longer periods of time.

As Chief Science Office Justin Kirkland explains, “The most novel part of this process is that we are performing two very different functions with a single unique component. This component works much like the attraction and repulsion properties of a magnet with a North and South end. We leverage this energy to perform two functions. First, the attraction energy will extract psilocybin out of the mushroom. Second, when this same material is applied to the skin, it will advance the drug through the skin and into the bloodstream.”

Containing all GRAS certified ingredients, this solution can then be loaded into our proprietary drug delivery system for direct passage through the skin into the bloodstream. Transdermal application avoids the first pass metabolism and degradation of psilocybin by the liver that occurs with traditional oral drug delivery, which increases bioavailability of the active ingredient.

In the continued effort to champion all things natural, Optimi’s delivery system has been designed to require no additional shuttling agents. Instead, the repulsion technology steadily pushes the active ingredient through the various layers of the skin, and treatment begins almost immediately upon application. This new process will help to reduce costs and resources while providing patients with a superior product.

The Company is in close contact with the regulator and welcomes finalization of the process in the coming weeks. Additionally, Optimi commends Health Canada for taking an active leadership role in finding solutions for the ongoing Canadian mental health crisis including access to psilocybin for therapeutic purposes through the Special Access Program (SAP).

Background

In Canada, drugs are authorized for sale only once they have successfully gone through the drug review process, including evaluation of safety, effectiveness and quality. However, Health Canada’s SAP allows practitioners to request access to drugs that are not available in Canada for the emergency treatment of patients with serious or life-threatening conditions who have exhausted other treatment options. Due to regulatory changes made in 2013, restricted drugs have not been made available through the SAP.

Since then, clinical trials have been the only avenue by which the sale of a restricted drug could be authorized for patient treatment. While clinical trials remain the best mechanism to authorize the sale of drugs, there are barriers to patient participation including geography or lack of clinical trial recruitment.

Yet, there has been ongoing scientific research into the efficacy and safety of certain restricted drugs. In response, Health Canada has today announced a reversal of the regulatory changes made in 2013 to Part C of the Food and Drug Regulations and has restored potential access to restricted drugs through the SAP.

What the regulatory amendments mean for Canadians

The regulatory amendments will allow physicians to request patient access to psychoactive substances, like psilocybin and MDMA, for psychedelic-assisted therapy. Decisions will remain on a case-by-case basis and will be reserved for serious, treatment-resistant or life-threatening conditions. The amendments have the potential to positively affect the lives of people experiencing serious mental health conditions and end-of-life distress when other therapies have failed, are unsuitable or are unavailable in Canada.

Earlier last year, Optimi received a Section 56 exemption for research purposes and this initial trial will be conducted at the Bloom Clinic, which is Calgary’s first psychedelic assisted therapy centre.
