However, a new University of Chicago study found no effect on mood or task performance after giving healthy volunteer 4 LSD microdoses 3-4 days apart in a placebo-controlled study.

Atrophy of neurons in the brain plays a critical role in pathophysiology of depression and related disorders. Increased structural plasticity in the brain neurons (mainly prefrontal cortex) has been linked to the sustained antidepressant effects of ketamine and other psychedelic compounds. BetterLife had previously shown the anti-depressant properties of BETR-001 in a rodent chronic variable stress model. The current study demonstrates that treatment of rat embryonic cortical neurons with BETR-001 increases the structural complexity of neurons (dendrite growth and complexity) and therefore, provides evidence of neural plasticity activity of BETR-001. In certain measurements of structural plasticity in neurons, BETR-001 performs better than ketamine in this model.

Fish on acid? Microdosing zebrafish with LSD shows its potential benefits for humans

New research suggests that microdosing doesn’t cause withdrawal symptoms. But can it ease withdrawals from other substances?

MacEwan University researchers found that zebrafish showed increased anxiety behaviour during withdrawal from alcohol, consistent with symptoms that humans experience.  

However, when the zebrafish were given microdoses of LSD for 10 days (by putting them in a “dosing tank” for a precise period of time), there were no observed behavioural changes. This indicates that microdosing is not addictive and does not cause withdrawal symptoms. 

A study that’s currently underway will examine LSD microdoses on alcohol withdrawal symptoms.

Update on Spinout Process

Blackhawk is also pleased to report that MindBio is well underway with its proposed spinout.

In December 2021, Blackhawk announced its intention to spinout MindBio as well as Digital Mind Technology, both wholly-owned subsidiaries of Blackhawk, into a newly formed subsidiary of spinco.  It is aniticipated that the spinco will ultimately pursue a separate public listing.

BETR-001 (2-bromo-LSD, formerly TD-0148A) is a non-hallucinogenic derivative of lysergic acid diethylamide (LSD). Previous published studies have not included any data on PK for BETR-001. It was also unknown if presence of food would affect the bioavailability of orally administered BETR-001. The current study conducted by contract research organization, Nucro-Technics (Scarborough, ON, Canada), demonstrated the following key results after a single dose of oral BETR-001 (capsule) administered to beagle dogs:

  • No significant difference was observed in bioavailability and total exposure of BETR-001 in PK profile of fed versus fasted beagle dogs.
  • Oral bioavailability (%F), defined as the fraction of oral administered drug that reaches systemic circulation, was calculated to be 61% and 63% for fasted and fed states, respectively (no significant difference).
  • The maximum systemic concentration (Cmax) of BETR-001 after a single oral dose was reached at 0.5 hr (Tmax), suggesting a quick uptake of the drug into the systemic circulation. The drug was detectable in systemic circulation eight hours post oral dose with an elimination rate constant (Kel) of 0.4 per hour, pointing to the fraction of drug eliminated per unit of time.

The findings demonstrate that oral administration of a single dose of BETR-001 can reach the therapeutic range in the systemic circulation. The PK elimination constant (Kel) of 0.4 per hour for BETR-001 indicates a low probability of toxicity as a result of drug accumulation in the systemic circulation.

MindBio recently received feedback from its Phase 1 clinical trial independent Data and Safety Monitoring Committee that the clinical trial is approved to continue after a successful six-month safety review of its use of LSD in healthy participants.  After its positive safety review, MindBio is progressing the development and enhancement of its microdose formulation of LSD in a form for commercial distribution and use.

The new microdose formula will be adapted in the Phase 2 clinical trials and assessed utilizing MindBio’s proprietary technology which will be used to determine an optimal microdose for patients.  

Health Canada Announces Significant Amendment to Special Access Programme

Blackhawk commends Health Canada’s leadership on their recent amendments to the Special Access Programme (SAP), which will now allow physicians to request patient access to psychedelic treatments, including psilocybin-assisted therapy.

The amendments under the SAP came into effect as of January 5, 2022, as published in the Canada Gazette, and allows physicians to request psychedelic treatments on behalf of their patients, on a case-by-case basis, for serious treatment-resistant or life-threatening conditions. This significant amendment has the potential to provide numerous Canadians struggling with serious mental health afflictions with alternative therapies where traditional treatments have failed, or where other treatments are unsuitable or are unavailable in Canada.

Canada’s SAP was designed with the purpose of allowing people access to new, potentially life-saving medications before they are formally approved for routine use in healthcare, however prior to today’s milestone amendment, psychedelic medications have been ineligible for Special Access Programme applications. The regulatory amendment will enable Canadian physicians to apply for psychedelic therapies in a similar process as to how other investigational medications are accessed prior to formal drug approval.

The amendment to SAP by Health Canada is an historic milestone in Canada’s approach to treating mental health conditions and signifies a growing recognition of the important and growing body of research that supports the efficacy of psychedelics and psychedelic-assisted therapies in treating mental health afflictions. According to Health Canada, access to psychedelic treatments will be provided where there is sufficient data to support the safety and efficacy of the targeted drug for a patient’s specific condition.

Ms. Gutte earned her bachelor’s degree of computer engineering from the University of Applied Sciences in Brandenburg, Germany. Ms. Gutte has had extensive experience as a controller and CFO of numerous publicly traded and private corporations in several industries.

The seeds of Morning Glory flowers contain a hallucinogenic compound that’s similar to LSD. It’s said to induce a dreamlike state and was even used in Mayan rituals to communicate with spirits.
